    Cng vi s pht trin ca x hi, khoa hc cng ngh th cm t ch phm sinh hc khng cn qu xa l vi mi ngi. Nhc ti ch phm sinh hc probiotic, prebiotic ta thy ngay c nhng li ch m chng mang li cho sc khe con ngi cng nh li ch trong chn nui, trng trt. Ch phm sinh hc ngy nay c ng dng rng ri trong nhiu lnh vc ca i sng nh y t, lm p, sn xut thc phm, s dng trong nng nghip. Tuy nhin, chng ta vn cha bit v hiu r c v ngun gc, quy trnh sn xut ca nhng ch phm . Vn ny s c trnh by trong ti Tm hiu quy trnh sn xut mt s ch phm probiotic v prebiotic. Trong phm vi mn Dinh dng hc, em ch tm hiu ti ny trn i tng ngi v c trnh by trong phn Ni dung bn di.

    1. CH PHM PROBIOTIC

    1.1. Tng quan

    1.1.1. Khi nim

    Probiotic l vi sinh vt sng trong khi c qun l ph hp v mt em li li ch cho sc khe trn vt ch (FAO 2001). C th hn, probiotic l cc vi khun sng c li ng rut, khi b sung vo c th em li cc li ch sc kho cho con ngi nh ci thin h vi sinh ng rut v chc nng ca rut.

    1.1.2. Cc th h probiotic

    Sn phm ln men t nhin: sa chua, da chua, ph mai, kim chi,c dng c li v mt dinh dng, nhng cha c tnh h tr tr liu mt cch r rng.

    Th h th 1, probiotic khng bao (non-coated): c cc sn phm nh: Antibio, Abio Gran, Biolac, Bio - Acimin, Bio - Baby,; vi khun c phn lp v tng s lng s dng, nhng vn l vi khun khng bao. Nhc im: d b hao ht khi bo qun v s dng.

    Th h th 2, dng bao tan trong rut (enteric- coated): vi khun cha trong nang c lp bo v ch tan trong rut.

    Th h th 3, dng bao vi nang (micro-encapsulated): nh Lactomin, gip vi khun qua d dy tt hn. Nhc im: tan chm trong rut.

    Th h th 4, bao kp (DuolacTM): nh Kidlac, Lacclean Gold, Chewable 7.1, Yamyam, probiotic c bao 2 lp, hiu qu cao nh kh nng sng cao n rut v ng thi khc phc nhc im phng thch chm rut nh cch bao cc th h c, nh lp bao th 2 (lp bao peptide) c tnh thc y phng thch ti ch.

    1.1.3. Li ch ca probiotic

    Probiotic c nhng li ch nh sau: tng cng kh nng tiu ha lactose v hot ng ca cc enzyme khc; tr tiu chy do khng sinh; tr vim ng tiu ha; c ch v tiu dit vi khun c hi; gim nguy c ung th rut kt; iu ha min dch, gim phn ng min dch gy vim; tng cng sc khe sinh sn ca ph n.

    1.1.4. Hot ng ca vi sinh vt probiotic

    Cnh tranh v tr dnh bm v ngun dinh dng vi vi sinh vt gy bnh.

    1.1.5. ng dng ca ch phm probiotic

    1.1.5.1. Trong y hc v thc phm

    S dng vi khun mui chua rau qu, chua thc n gia sc (lm chn sinh hc cc loi qu): To c sinh khi vi khun c ch, t c sinh vt gy thi. Gy chua, to hng v thm ngon cho sn phm. Chuyn rau qu v dng chn sinh hc do m hiu sut tiu ha tng. Mt s chng dng sn xut sa chua c: Lactobacillus bulgaricus, Strepticoccus faecalist Mt s chng c thm vo sa bt: Lactobacillus, Bifidobacterium Ti nhng nc pht trin, cc nh sn xut, ch bin ng dng b sung Probiotics vo nhiu loi thc phm nh sa chua n, phomt, kem.

    Cha bnh ng rut: Php sn xut v a ra th trng t hng chc nm nay mt sn phm mang tn Biolactyl chuyn tr tiu chy bng nhiu khun Lactic. Vt liu sinh hc.

    1.1.5.2. Trong nng nghip

    Probiotic c ng dng rng ri trong nng nghip: sc sn, thy sn, trng trt Probiotic gip ci thin sc khe ca ng vt, gip tng trng, gim t l cht non v ngn chn tc nhn gy bnh. S lm dng khng sinh trong ngnh chn nui v kh nng khng khng sinh lm tng mi quan tm n probiotics.

    1.1.6. Cc dng ch phm probiotic

    Probiotic c lnh (yophilis): khi h nhit xung tht thp vi khun s tr nn kh nh bt nhng khng b tiu dit. Chng s sng tr li khi c a vo c th.

    Vin nang (capsule): thng c hai hoc bn loi phi hp vi nhau nh L.casei, L.acidophilus, L.bulgaricus, L.rhamnosus, Streptococcus thermophi- lus, B.lactis, B. longum.

    Bt (poudre): dng bt pha trong nc. Nh lu li bao t trong thi gian ngn nn khi n rut, mt s lng ln vi khun vn cn sng.

    Sa chua tr liu: Nn ung trc ba n, c th cha mt t l probiotic rt cao (50 t CFU/ liu).

    1.2. Sn xut ch phm probiotic

    1.2.1. Chn la vi sinh vt probiotic

    Hin nay, cc ch phm probiotic ang c sn xut nhiu u ch yu cung cp cho ng vt nui, gia sc, gia cm ring i vi ngi th ang rt hn ch. V nhng ch phm s dng cho ngi nh hng trc tip n sc khe con ngi, vic sn xut cc ch phm ny

    cn phi lun cn thn v ch tng giai on. V vy khi sn xut trc ht cn chn ng loi vi sinh vt, sau cn kim sot cht ch cc yu t c th nh hng n cht lng sn phm.

    Khi chn la vi sinh vt probiotic cn quan tm n cc yu t sau: kh nng bm dnh ln thnh rut; khng sinh c t, khng gy hi vt ch; to sn phm vt ch s dng c; c kh nng sinh trng sinh sn tt; d nui cy, s lng t bo qun th ln; chu c pH thp; sng st c khi bao gi (bo t); nhit .

    1.2.2. Quy trnh sn xut tng qut Di y l quy trnh sn xut tng qut ch phm probiotic dng kh v dng lng.

    Chun b ging

    Chng vi sinh vt sau khi c tuyn chn, thun ha gip vi sinh vt tng kh nng thch nghi vi iu kin thc t v lnh vc phc v. Tip theo, chng vi sinh vt c em i nhn ging to thnh ging s dng cho sn xut. H thng lu gi ging vi sinh thun chng phi c bo qun theo tiu chun k thut c bit. Cn kim tra mt v trng thi hot ng ca vi sinh vt. Cc thng s phi c ghi nhn v bo co hng gi.

    Nhn ging

    Mi trng dch th chun b cho qu trnh nhn ging m bo y cc cht dinh dng, cc nguyn t a lng v vi lng. Trong mi trng ny, vi sinh vt sinh trng, pht trin nhanh chng v dn n nh thch nghi trc khi c a vo quy trnh ln men. Nhn ging cn m bo to ra v duy tr ngun ging thun chng mang y cc u th ca ging gc.

    Dch men chun b c em i ng kh to khi vi sinh vt. Khi vi sinh vt ny c nghin thnh dng bt, c nng vi khun cao. Sau bt c trn, nn. Cui cng men vi sinh c ng gi trong nito, nhm mc ch bo qun, ta c thnh phm.

    1.2.4. Mt s ch phm probiotic

    1.2.4.1. Ch phm Viabiovit

    Viabiovit c thnh phn l cc vi khun lnh tnh h Lactobacillus (Lactobacillus sprorogenes, Lactobacillus kefir, Lactobacillus acidophilus) l thnh phn chnh v cng quan trng v khng th thiu trong cc ch phm men vi sinh kt hp vi cc vitamin nhm B v vitamin PP c tc dng tng cng kh nng tiu ho v hp thu tt cc cht b dng cho tr. Sn phm c phi hp 3 chng vi khun trong h Lactobacillus rt c li cho ng rut. Vi hm lng vi khun ln v dng ng kh nn kh nng sng bo tn lu khi c bo qun nhit thng. Cc chng vi sinh vt ny s gip cn bng h thng vi khun c ch trong ng rut, gip thc y qu trnh tiu ho v hp thu dinh dng.

    C ch tc dng ca Viabiovit: lm tng cng hp thu thc n nh cc hot tnh ca 3 chng vi khun c ch.

    - To mi trng axit nh tit ra axit lactic, y l mi trng trng thun li cho h vi sinh ln men ng rut pht trin v ngc li, c ch s pht trin ca vi khun gy bnh dng Gram(-).

    - Tit ra cc cht dit khun nh: acid acetic, acid benzoic

    - Thc y qu trnh tng hp vitamin v mt s acid amin cn thit cho c th, do rt c li cho qu trnh tiu ho v hp thu.

    - Tc dng ca mt s vitamin nhm B v PP: nng cao sc khng ca c th, c bit gip tng cng kh nng tiu ho v hp thu tt cc cht b dng cho tr s sinh.

    - Cn bng ri lon h vi khun ng rut do dng khng sinh. Gip to cm gic ngon ming v tng cng tiu ho, dng tt cho nhng ngi ri lon tiu ho (tiu chy, to bn, i trng mn tnh, y hi) khng ch vi tr em m cn c tc dng tt vi nhng ngi c nguy c bnh tim mch v chng lo ho tui gi.

    1.2.4.2. Ch phm Kidlac

    Ch phm Kidlac l ch phm vi sinh dnh cho tr em. L s kt hp synbiotic gia 4 chng probiotic vi prebiotic s dng cng ngh probiotic bao kp. Sn phm thuc th h probiotic th 4. Probiotic c bao 2 lp, hiu qu cao nh kh nng sng cao n rut, c tnh thc y phng thch ti ch nh lp bao th 2. Probiotic nh lp bao polysaccharid gip bo m sng tt trong qu trnh sn xut v bo qun, lp bao protein c c tnh ng vn trong mi trng acid gip chng c tc ng khi qua dch d dy v mui mt n rut. Ti rut, nh lp protein c phn tn tt trong pH trung tnh hoc hi kim rut, probiotic s phng thch tt nh sn sinh nhanh chng pht huy ti a tc dng.

    Kidlac c phi hp nhiu chng, c tc dng tt hn dng ring l. Kidlac l s kt hp ca bn chng vi khun: Lactobacillus acidophilus (100 billion cfu/g), Bifidobacterium longum (50 billion cfu/g), Bifidobacterium breve (50 billion cfu/g), Enterococcus faecium (100 billion

    cfu/g).

    Hiu qu khi s dng Kidlac:

    - Gim nhanh chng cc ri lon ng rut do bnh l v/hoc dng khng sinh ko di hoc nhim nhng loi khun gy hi, thuc ha tr liu, gy nn cc triu chng nh tiu chy, vim rut, chng bng, y bng, i phn sng, ri lon tiu ha, tng s thi ra trong rut.

    - Ci thin cc tnh trng mn tnh do ri lon ng rut thng xuyn v lu di nh: to bn, vim i trng, thng xuyn b trc trc tiu ha v tiu chy, b tiu chy khi ung sa do khng dung np lactose

    - Ti lp cn bng t nhin trong h tiu ha, kch thch s pht trin ca cc vi khun c li cho sc khe, gip ng rut khe mnh.

    - Gip gim nhim khun v d ng i vi cc khng nguyn c trong thc phm.

    - Tng kh nng hp th dinh dng, cung cp cc vitamin cho c th nhm nng cao sc khe, tng sc khng ca c th.

    - Gip tng cng kh nng min dch, phng nga v khng nhiu bnh khc nhau.

    - Cung cp cc vitamin cho c th nhm nng cao sc khe, colostrum gip tng kh nng min dch v DHA gip pht trin tr no.

    2. CH PHM PREBIOTIC

    2.1. Tng quan

    2.1.1. Khi nim

    Prebiotic c nh ngha nh l mt thnh phn thc phm khng tiu ha c tc ng c ch n vt ch bng cch kch thch c chn lc s pht trin hoc hot ng ca mt hay mt s lng gii hn cc loi vi khun trong rut kt v do gip ci thin sc khe ca vt ch (Gibson v Roberfroid, 1995). Prebiotic ch cc loi cht x ha tan c mch ngn, ch yu l cc hp cht oligosaccharide.

    2.1.2. Li ch v tc hi ca prebiotic i vi sc khe

    Prebiotic l ngun thc n cho probiotic, to iu kin cho li khun pht trin mnh m v ci thin h tiu ha ca c th. Prebiotic khng b tiu ha d dy, rut non nn n cn nguyn khi xung rut gi, prebiotics nui dng vi khun rut gi v lm tng hot ng ca chng y.

    Prebiotic tng cng hp thu canxi, magie v cc cht khong khc; nng cao v tng cng h min dch ca c th; gim nguy c ung th trc trng v ngn nga ung th rut kt giai on u; duy tr mc ng huyt, h tr ngi bnh tiu ng, gim cholesterol xu, h tr n nh huyt p, duy tr thoi mi cho ng rut, ngn chn s xm nhim ca mt s vi khun gy bnh, chng su rng, gim nguy c d ng tr em, h tr tiu ha d dng hn v hn ch cc trng hp kh tiu do y hi, to bn.

    Tuy nhin, prebiotic ch c tc ng tch cc ti c th c sn cc vi sinh vt hu ch, bn thn prebiotics khng sinh ra cc vi sinh vt ny. S dng nhiu prebiotics gy tnh trng y hi, i tiu nhiu hn 2 ln/ngy. Cc loi ng c ngun gc t FOS c th kch thch s tng trng ca vi khun Klebsiella l mt vi khun gy bnh ng rut. Tiu th prebiotic lm tng vi khun to kh gas trong h tiu ha, do cc sn phm mi pht trin gn y thng hng ti vic y mnh s pht trin ca vi khun khng to kh. Prebiotic c tc dng khc nhau ln cc i tng khc nhau. V d FOS khng nh hng n s hp thu canxi thanh nin nhng li kch thch s hp thu ny thiu nin. V vy cn ch khi s dng prebiotics.

    2.2. Sn xut ch phm Prebiotic

    Prebiotic c sn xut ch yu bng bn cch:

    - Chit tch trc tip t nguyn liu th thc vt nh: inulin t rau dip xon, oligosaccharide soybean t u tng

    - Thy phn c kim sot: thy phn c kim sot polysaccharide bng enzyme, c th s dng phng php sc k tinh sch prebiotic nh fructooligosaccharide t inulin.

    - Glycosyl ha: qu trnh tng hp bng cch s dng enzyme thu phn to oligosaccharide t cc disaccharide v c tinh sch bng qu trnh sc k nh GOS t lactose, FOS t sucrose

    - Cc qu trnh ha hc: s chuyn i xc tc t cc cacbohydrate nh Lactulose t s ng phn ha lactose trong mi trng kim.

    2.2.1. Sn xut FOS

    2.2.1.1. Gii thiu v FOS

    FOS (fructooligosaccharide) l cht x ha tan, l cht to ngt nng lng thp. FOS kch thch tiu ha, gim cholesterol, phospholipid, triglyceride, chng bo ph, an ton cho ngi bnh tiu ng, nga su rng, tng kh nng hp th cc ion Ca2+, Mg2+.

    FOS l nhng oligosaccharide c 1 phn t sucrose lin kt vi 1, 2 hay 3 gc fructosyl

    bng lin kt 2,1 glucoside.

    Syrup FOS thng mi gm 25 30 % 1 kestore (GF2), 10 15 % nystose (GF3) v 5 10 % 1

    F- fructofuranosylnystose (GF4) v 25 30 % glucose nh mt sn phm ph (Kim v cng s,

    2000).

    2.2.1.2. Quy trnh sn xut FOS trong cng nghip

    Trong hai phng php trn, phng php lin tc c nhiu u im ni tri v kh nng cng nghip ha cao hn.

    2.2.1.3. Cc yu t nh hng n sn xut FOS

    Loi vi sinh vt thng s dng lm ngun enzyme sn xut FOS l A. niger, Auerobasidium pullulans v Auerobasidium japonicus (tt nht sn xut FOS loi 1F)

    Enzyme t nhng ngun khc nhau thng c khong dao ng t 232 kDa (Imamura v cng s, 1994) n 246 kDa (Hayashi v cng s, 1991).

    Qu trnh sn xut FOS cn nh hng bi cc yu t nh: pH, nhit . pH ti u sn xut FOS l t 5 n 6, nhit ti u t 50oC n 60 oC. Nhng gi tr ny c th thay i ty theo ngun enzyme. Nng sut ti a ca FOS th khc nhau theo ngun v phn ng c thit lp, t 42% (Kim v cng s, 1998) t Bacillus macerans n 80% (Madlova v cng s, 1999) t Auerobasidium pullulans v A. niger.

    Mc d phng php sn xut FOS tn ti c thit lp rt tt nhng nhiu nh nghin cu vn tip tc tm ra cch lm tng nng sut v hiu qu ca phn ng ti quy m cng nghip, cng nh cc phng php n gin ha v thc y phn ng ca chnh n.

    Ngun enzyme khc nhau v iu kin phn ng c s dng trong cc n lc ti u ha k thut c nh enzyme.

    Chien v cng s (2001) hon thnh sn xut FOS qua vic c nh t bo ca A.japonicus ti gluten trong phn ng gin on. Mt gam si c c nh c vi 100ml ca 400g l-1 dch ng saccharose. Lm tng 61% nng sut sn xut FOS sau 5 gi, sn phm gm ch yu l 1 kestose v nystose. S tp trung ca saccharose v t bo trong gluten c nh hng ng k n tc phn ng ca enzyme.

    2.2.2. Sn xut GOS

    2.2.2.1. Gii thiu v GOS

    GOS (galactooligosaccharide) l mt prebiotic c ngun gc t ng vt, c chit sut t lactose c trong sa b, d. Mt s GOS c s dng nh mt loi cht to ngt nng lng thp, thnh phn ca thc phm v cht thm vo m phm.
