Amylomaltase (EC 2.4.1.25) ftom Corynebacterium glutanicum is a gl kDa proteinwhich belongs to the 4-o-glucanotransferase (4ocTase) family. This enzymecatalyzes the tansfer of o-1,4-D-glucan units via an intemolecular andintramolecular transglucosylation reaction to produce longer chain oligosaccharidesand cycloamyloses, tespectively. In order to understand the mechanisms of Cglutdmicum amylomaltase (CgAM) at molecular level. the three-dimensional strucluleanalysis is essential. The CgAM has been cloned and exprcssed in Escherichia ColiBL21(DE3)'. However, the amino acid sequence of C.gAM is 28oZ and 33oZ simlar rothose of amylonaltases ftom Thermus aquaticus, rlird, Solanum tube osums . of whicbtheir crystal structutes have already been solved. Crystallization of CqAM has beenobtained by the hanging-drop vapour-diflusion nretbod. preLiminary d;1a shows thatthe crystal diffiacts to 2.5 A at synchrotron source INSRRC. Taiwan). However, themolecular-replacement method does not provide corect solution for strucruredetermination. Therefore, tlte selenomethionine (Se-Met) substituted protein crystalsare rcquired. The C'gAM was expressed in E colt 8834(DEj) that is a MethionineAuxotroph strain. We found that this enz),rne displays normal disporportionauonactivity on maltotriose substate. The maxjmunr specific acri\,ity is oblain;d when .lerecombinant cell was cultured at 37 oC for 4 hours after induced with 0.4 mM IPTG.Pudfication of Se-Met CgAM is now in progress.

The biochemistry, enzymology and molecular biology are interesting andimportant fields for life science development today. The scope of these fields alsooffers significant intellectual stimulation and challenge. These concems have mademe as a bachelor in Biotechnology Aom King Mongkut's Institute of TechnologyLadkabang choose the department of Biochemisty, Faculty of Science,Chulalongkom University for my gmduate studies. I have joined the "Starch andCyclodextrin Research Unit" Laboralory which is specialized in the reseaxch area ofstarch metabolism, cyclodextrin producing and carbohydmte-modifying enzymes.

My Ph.D. rcsearch focuses on the molecular studies of bacterialamylomaltase, a large- ng cyclodextrin producing enzyme. Cloning, characterizationof the enzyme and site-directed mutagenesis for identifying residue involved inenzyme catalysis were performed. In my 1" year of Postdocteral training, expressionof the enzyme in yeast which is a GRAS organism for applicable use in food hasbeen focused. Due to the potential advantage of amylomaltase in industry, mypresent work involves stuctural determination ofthe enzyme by x-ray crystallizationto disclose basic knowledge in this enzyme which will lead to more understanding ofits mechanism. To proficient in an)'thing, it requires extensive study and endlesspractice, sometimes even life-long.

In my opinion, the Youlg Scientist Progran is a meaningful program foryoung researcher in Biochemistry and Molecular biology areas. It is a geatopportunity for many ofthe young PhD students or Postdocs to present their researchworks and discuss about the new findings, sharing their lab experiences andknowledge to each other. Not ody an academic program, the participants will have achance to have activities together which might provide collaboration and networkingin the future. Finally, I believe the YSP is the right specialty for me and I am excitedto participate in this program.
