II. 균주 ( cells or spores )
1. 균주의 획득
1) 자연계에서 분리
① Sources : 대기, 토양, 특수지역, 식물 및 동물세포
② 미생물의 순수 분리
- Continuous enrichment culture (연속농화배양)
- Plate culture (단일 colony분리를 반복) : 선별배지의 이용
③ 새로운 metabolites를 얻기 위한 미생물 탐색
- 새로운 미생물의 탐색 : 30 ∼40%
예 1) 꿀 벌집의 삼투압 내성 효모 : 당알콜 생성
예 2) Sea and fresh water의 Actinoplanes strains :
20,000 strains 중에서 13,000 strains 가 항생제 생성
⇒ 41개의 새로운 항생제 분리

2. 균주의 관리보존
1) 균주 보존시 유의점
① 보존중의 사멸이나 변이 방지
② 보존기간을 가능한 한 길게
③ 접종원으로 반복해서 사용할 수 있도록
④ 오염되지 않도록
⑤ 조작이나 용기가 가능한 한 간편하게
2) 균주의 보존방법
① 계대배양법: Slnat에 배양하여 보관 (4℃, 55% 습도, 2 ∼ 6개월)
② Test tube 배양법 : 액체배양 후 밀봉하여 저온 보관
③ 냉동보존법 (Deep freezing method) : 장기간 보존이 가능
- 액체배양 후 영하180℃에 보존 (액체 질소에 의한 급속냉동)
- ice crystal이 형성되지 않도록 cryoprotectants 이용
• Bacteria : glycerol, ethylene glycol
• Yeast, Fungi, Plant & Animal cells : Dimethylsulfoxide
④ 냉동건조법 (Lyophilization) : 미생물의 장기간 보존방법
미생물 배양 → 배지에 현탁 → 급속냉동 → 냉동건조 → N2 gas 주입
→ ample 보관 → 다시 이용시에는 멸균증류수에 넣어 배양
3. 균주의 개량
1) 목적 : 경제성
① 균주의 산물생성 향상
- 수율(고가의 원료), 생산성(배양기 용량), 최종농도(분리정제)
② 발효특성 개선
- 부산물의 생성 억제 (분리정제 용이)
- Foam 생성, 산소요구량, 낮은 viscosity (통기 교반에 용이)
- 값싼 기질 이용과 고농도 탄소원과 질소원에 내성
- Phage에 저항성인 균주 (phage 오염방지),
예) Wako 등(1998) ; 산업적인 생산에 이용
- 곰팡이인 Aureobasidium sp. 가 40%의 수율로 erythritol 생산
⇒ 균주개량에 의한 foam 생성제거에 의하여 45%로 수율향상
2) 돌연변이(Mutation)
① Mutagens
- Physical mutagens : UV, γ-ray
- Chemical mutagens : HNO2, alkylating agents (EMS, NTG 등),
Acridine orange (frame-shift)
② 변이유도 조건
- mutagen 처리에 대한 strain specificity
예) Bacteria : HNO2, UV
Yeast, Eukaryote cell : EMS, NTG, UV, γ-ray 등
- mutagen 처리 조건 : pH, buffer composition, mutagen 농도,
미생물의 growth phase, 처리시간 등

③ 균주개량 원칙
- 서로 다른 mutagen을 처리
- step by step으로 수행
- 원하는 배지 조성 및 배양 조건에서 test할 것
④ 변이균주의 선별
Random selection : 직접 metabolite나 발효특성의 측정에 의한 선별
- 일의 양이 많지만 현재까지도 산업체에서 균주개량에 많이 이용
- 계속적인 mutation과 selection을 가장 좋은 strain을 가지고 반복하여
균주의 생산능력을 점차 증가시킴

3) Genetic recombination
- 서로 다른 두개의 genotype을 재조합시켜 새로운 genotype을 갖는 strain으로 개발
① 생성물은 증가시키고 대립 유전자가 하나의 strain에 존재할 수 있으므로 유전자의
누적효과에 의하여 high yield strain을 얻을 수 있다.
② Mutation에 의하여 생산성 증가가 어려운 경우 mutant와 parent간의 recombination
에 의하여 생산성이 증가되면서 발효특성이 개량된 strain을 얻을 수 있다.
예) Fungi의 sexual 또는 parasexual recombination
4) 세포융합 (Protoplast fusion)
① 모든 세포를 이용한 genetic recombination이 가능
② 방법 : 원형질체 형성 → Ca++, PEG 존재하에서 융합 → cell wall regeneration
→ 선별
5) 유전자 조작 기술
① 균주개량 : Metabolic pathway에서 rate limiting step의 유전자 증폭
② 외래 유전자 주입 재조합 미생물 : 외래 단백질의 생산 (재조합 단백질 의약품 등)
☞ 외래 유전자 이용시 고려사항
- Foreign DNA : cDNA, synthetic DNA, PCR , codon usage, mRNA의 stability,
ribosome과의 affinity
- Expression vector : copy number, selection marker, stability
- Promoter : regulation (repressible & constitutive), strength
- Host cell : low protease, GRAS (generally regarded as safety), auxotroph, etc.
- Secretion : signal peptide, glycosylation, proteolytion.
6) 대사공학 기술
대사과정의 흐름(metabolic flux)을 분석하여 원하는 방향으로 재조정하는 기술
① metabolic flux 분석 : DNA chip 이용
② 균주 개량 : 유전자 조작과 단백질 공학 기술 이용
③ 이용 범위
- 대사경로의 활성화에 의한 산물의 과량생산
- 기질의 이용범위 확대
- 유해 물질에 대한 저항성 향상
- 새로운 물질의 생산 능력 부여
- 균주의 배양조건 특성 변형 등

Ⅳ. Bench-scale fermenter (1 ~ 5 L)
1. Inoculum 준비 (5 ~ 10%, v/v)
1) 접종용 배지의 선정기준
① lag phase를 최소화
② 균주가 health하고 세포성장이 우수해야함
③ 대량 volume의 배양이 가능
④ 균주의 morphological form이 적당해야 함
⑤ contamination이 없는 원료 이용
⑥ 균주의 생성물 생성 능력이 유지되어야 함
2) 접종용 균주의 배양조건, 세포농도, 배양시간 결정
2. 생산용 배지조성 최적화
☞ 세포성장, 산물생성, 에너지 공급 및 생명 유지에 이용
1) 산업용 배지의 선정 기준
① 산물의 생산수율과 최대 농도 고려
② 부산물 생성을 최소화
③ 저렴한 가격과 년중 구입이 용이
④ 생산공정상의 문제점 최소화 (통기 및 교반, 분리정제 과정, 폐수처리 등)
2) 물 (Water)
① 가장 많은 양이 소모되므로 permanent하게 공급이 필요
② pH, solids양, contamination 정도, mineral content 등의 확인이 필요
3) 탄소원 (Carbon source)
① Glucose & sucrose : 가격이 비싸다
② Molasses : 아미노산 및 핵산 발효에 이용
③ Hydrol : 액상임
④ Malt extract : 곰팡이, 효모, 방선균에 좋은 기질임
⑤ Starch & dextrin : amylase 생산, ethanol 생산
⑥ Sulfite water liquors : yeast 배양에 이용
⑦ Cellulose : 볏짚, 옥수수속, bagasse 등
⑧ Whey : lactose로 이용
⑨ 동물 및 식물 기름 : 기질 보조제로 많이 이용
⑩ Methanol : 가격이 가장 저렴하지만 이용할 수 있는 균주가 제한
⑪ Ethanol : 식초생산 원료
⑫ Alkanes : 원유가격 상승으로 이용 가능성이 희박
4) 질소원 (N-Source)
① Chemical compounds :
(NH4)2SO4, urea, nitrate salts, 암모니아수(과량 첨가시에 미생물 생육이 저해됨)
② 유기 질소원
- Light steep water (LSW), corn steep liquor (CSL), corn steep powder (CSP)
- Yeast extract : 가장 우수하지만 가격이 비싸다.
- Peptones (protein hydrolylate)
- Soy meal : 항생제 발효에 많이 이용
5) Minerals
① 기본적인 minerals : Mg2+, Pi, K+, Ca2+, Cl-
② Trace elements : Co, Cu, Fe, Mn, Mo, Zn 등
6) Vitamin source
① 기본적으로 C-source와 N-source에 존재
② Cofactor로 꼭 필요한 경우에는 첨가
7) Precursor 및 metabolic regulators 첨가
예) Penicillin → Phenylacetic acid, Enzyme → inducer
8) 산소(호기성) : 교반과 통기에 의하여 제공
9) Antifoam : foam 발생시 첨가
내압 (0.1 ~ 0.4 bar) - foam 제거 및 KLa 증가
3. 배양조건 최적화
☞ 산물의 수율, 생산성 및 최종 농도를 고려하여 설정
1) 배양 pH : Non-control 또는 control
- CaCO3 첨가 : 산 생성에 의한 pH 저하 방지
- Phosphate 및 protein, amino acid : buffer 능력이 존재
2) 배양 온도 : 균주에 따라 다양하여 세포 성장과 산물의 생성조건이
다를 수도 있음
3) 통기 (Aeration) : 호기성 미생물에서는 세포성상에 매우 중요
세포내 oxido-reduction potential에도 영향
4) 교반 (Agitation) : 방선균 및 곰팡이 에서는 morphology와 산물의
생성에 큰 영향을 줌
4. 배양 방법
1) 회분식 배양 : 높은 수율 및 최종농도, 생산성이 낮음
2) 유가식 배양 : 높은 최종농도와 생산성
① Intermittent feeding 방법
② Continuous feeding 방법 (constant, linear, exponential)
③ pH-stat
④ Glucose-stat
⑤ Two-stage (growth stage → production stage)
3) 연속배양 (continuous culture) : 가장 높은 생산성, 최종산물의 농도가 낮음
고정화 세포의 이용에 가장 적합함
5. Scale-up
1) Scale-up 단계
① Bench scale : slant → 500 mL flask
② Pilot plant scale : 5 L → 50 L → 500 L
③ Production scale : 5,000 L → 50,000 L∼100,000 L
2) Scale-up의 basis
① Constant volumetric power imput (Pg/v)
알코올과 유기산 발효에서 발효조의 geometric similarity와 Pg/v로 scale-up
② Constant volumeteric oxygen transfer coefficient (KLa)
산소요구량이 높은 항생제 발효에서 scale-up에 이용
③ Constant impeller tip speed (πDii)
곰팡이의 morphology 유지 (pellet)
④ Constant mixing time (t )

2. Ornithine 생산 (Scale-up)
1) 균주 : Brevibacterium ketoglutamicum (glutamic acid 생산균주)
⇒ L-citrulline auxotroph이면서 arginine hydroxamate 내성 변이 균주
