Ninhydrin

Ninhydrin (2,2-Dihydroxyindane-1,3-dione)

is a chemical used to detect ammonia or

primary and secondary amines. When

reacting with these free amines, a deep blue

or purple color known as Ruhemann's

purple is evolved. Ninhydrin is most

commonly used to detect fingerprints, as

amines left over from peptides and proteins

(terminal amines or lysine residues)

sloughed off in fingerprints react with

ninhydrin.

Protein

• Proteins are complex, specialized

molecules composed of carbon,

hydrogen, oxygen, and nitrogen. Many

proteins also contain sulfur. The building

blocks of proteins are the amino acids.

* Rack tube

* A test tube

* Pipette drops

*Heater

* Tube clamps

* Albumin 2%

*Gelatin 2%

* Pepton 1%

*Ninhydrin reaction 0,1%

dry, 2ml subtance test

* Added 5 drops of ninhydrin

*Then, burn on during ±5 minute

CONCLUTION

From the results of an experiment in getting results :

• Albumin + ninhydrin solution + heat purple

• Gelatin + ninhydrin solution + heat purple

• Pepton + ninhydrin solution + heat purple

So the solution is a positive reaction is Albumin, Gelatin

and Pepton.
