1. Introduction The aim of the validation study is to evaluate the performance of the SMS method regarding the reference method EN ISO 6579 for the categories of products defined by the scope of application according to the standard ISO 16140:2003 and its amendment A1:2011.
1.1. Validation history The validation history of the method is the following:
‐ May 2004 : initial validation of the method according to the standard ISO 16140:2003, ‐ July 2007 : extension of the method for addition of the SMS confirmation tests to confirm the presumptive positive results, ‐ March 2008 : first renewal, ‐ March 2012 : second renewal, ‐ May 2016: third renewal
1.2. Alternative method The SMS test is designed to detect motile Salmonella spp. The test kit allows the presumptive detection of the pathogen target bacteria after a minimum of 16 hours of enrichment when present in the sample. The protocol of the method is showed in figure 1.
Pre‐enrichment 25 g sample in 225 mL buffered peptone water (1/10th dilution)

Principle of the assay SMS is a simple and economical rapid test for Salmonella spp detection in foodstuffs. The principle of this method is based on the motility of Salmonella and its Lysine decarboxylation activity. Salmonella spp produce migration areas associated with a change of the colour of the media. Incubation temperature of the media (41°C) gives more selectivity to the SMS method. The SMS test doesn’t detect the non‐motile Salmonella spp.
1.3. Reference method The standard EN ISO 6579:2002: Horizontal method for the detection of Salmonella spp has been applied. The protocol of this method is presented in figure 2.

1.4. Validation repositories The method has been validated according to the standard EN ISO 16140/A1:2011: Microbiology of food and animal feeding stuffs ‐ Protocol for the validation ‐ of alternative methods and to the specific requirements of the Technical Board linked to this standard.
1.5. Scope of application The alternative method was tested for all food and feed products and environmental samples.

2. Methods comparison study The following parameters were studied during the comparative study:
Relative accuracy (AC), relative specificity (SP) and relative sensitivity (SE)
Relative detection level of the alternative method and the reference method
Selectivity of the alternative method
Practicability of the alternative method
2.1. Relative accuracy, relative specificity and relative sensitivity The relative accuracy is the degree of correspondence between the response obtained by the reference method and the response obtained by the alternative method on identical samples. The relative specificity is the ability of the alternative method to not detect the target microorganism when it is not detected by the reference method. The relative sensitivity is the ability of the alternative method to detect the analyte when it is detected by the reference method. The objective of this study is to evaluate the performance of both methods on contaminated and non‐contaminated samples.
2.1.1. Initial validation
2.1.1.1. Number and nature of samples
The following categories were studied: meat products, dairy products, seafood and vegetable products, egg products, feedstuffs and environmental samples. A number of 378 samples was analysed. Types of products are indicated in table 1.

2.1.1.2. Artificial contamination of samples
Artificial contaminations of food samples were performed. For spiking, several strains were stressed using different treatments and the stress intensity was evaluated (logarithmic difference between enumeration on non‐selective agar –TSA‐ and selective agar –XLD‐). Stress values are shown in appendix 1. Among positive results, samples spiking was realized on 138 samples so 25.5% of naturally contaminated samples were analyzed.
2.1.1.3. Confirmation protocol
For alternative method, confirmation of presumed positive samples was performed from SMS media. A streaking on XLD and HEKTOEN agar was realized. After incubation of the media in adequate conditions, typical colonies were isolated on nutrient agar and analyzed by biochemical and serological tests. In 2007, an extension study was performed to add new protocols of confirmation: 150 target strains (Salmonella spp) and 105 non‐target strains were tested. Two protocols were proposed: a five hours protocol using a latex test and a 24 hours protocol using the SALSA medium and a latex test. The results showed good concordance with the two protocols.
2.1.1.4. Results
Each sample was analyzed once by the alternative method and once by the reference method. Table 2 presents paired results of both methods. Raw results are shown in appendix 2.

For all products categories, these results allowed calculating the relative accuracy, relative specificity and relative sensitivity according to the standard EN ISO 16140:2003. Results are indicated in table 3.

2.1.1.6. Analysis of discordant results
Discordant results are examined according to annex F of NF EN ISO 16140 standard, with Y as the number of discordant results. This analysis is presented in table 6.

The SMS test and the reference method EN ISO 6579 can be statistically considered as equivalent.
Negative deviations For one sample, the reference method gave a positive result while the alternative method result was negative. The confirmation tests showed the presence of S. Brandenburg.
Positive deviations For three samples, the alternative method gave a positive result while the reference method result was negative. All these results were confirmed by serological and biochemical tests. S. Enteritidis (seafood product) and S. Dublin (dairy product) were observed. Remark: for the positive results, the MUCAP test (confirmation test) was applied and the results were positive.
2.1.2. Third renewal In order to fulfil the specific requirements of the Technical Board linked to the standard ISO 16140:2003 for the validation of alternative methods, all samples with contamination levels superior or equal to 30 CFU/25 g were re‐tested using the protocols described in the standard ISO/FDIS 16140‐2:2015. This operation concerned 32 samples distributed among 5 categories. Statistical tests and interpretation of the results were still performed according to the standard ISO 16140:2003. Samples with a contamination level higher or equal to 30 CFU/25g from the initial study and the new sample tested in the 2016 study are shown in Appendix 2 bis.
2.1.2.1. Number and nature of samples
A number of 54 samples was re‐analyzed among which 34 were positive by any method. The new total of samples analyzed is 432. Types of products are indicated in table 7.

2.1.2.2. Artificial contamination of samples
Artificial contaminations were performed for all the samples added. A contamination by spiking was performed for two dry products of the category “Feedstuffs”. A contamination by seeding was performed for all the other products. Samples were inoculated and stored two to three days at 5±3°C. All stresses and contamination levels are mentioned in appendix 2 ter. Among all positive results, samples spiking was realized on 172 samples so 21.5% of naturally contaminated samples were analyzed.
2.1.2.3. Confirmation protocol
For alternative method, confirmation of presumed positive samples was performed from SMS media: ‐ by using the 5‐hour and the 24‐hour protocols of SMS Confirmation,
‐ by using the protocol of the reference method after streaking on XLD and ASAP: after incubation of the media in adequate conditions, typical colonies were isolated on nutrient agar and confirmed by biochemical and serological tests.
2.1.2.4. Results
Each sample was analyzed once by the alternative method and once by the reference method. Table 8 presents paired results of both methods. Raw results are shown in appendix 2 ter.

The SMS test and the reference method NF EN ISO 6579 can be statistically considered as equivalent. Two negative deviations appeared on two dairy products during these new experimentations:
‐ sample SMS 23: an atypical profile was found on the SMS agar media and no typical colony was able to be found from a streaking from the SMS agar plate. For this sample, it may be possible that the concentration in Salmonella in the enrichment broth didn’t reach the threshold of the method as the contamination level was very low (0.4 CFU/25 g). ‐ sample SMS 42: an atypical profile was found on the SMS agar media. Typical colonies were found from a streaking from the SMS agar plate. For this sample, it may be possible that the high level of annex flora masked the presence of Salmonella on the SMS agar media or inhibited its growth.
2.2. Relative detection level The objective of this study is to determine the level of contamination for which less than 50% of the responses obtained are positive and that for which more than 50% of the responses obtained are positive.
2.2.1. Initial validation
2.2.1.1. Matrices
Five couples “matrix‐strain" were studied in parallel with the reference method and the alternative method for all categories. The total viable count of each matrix was enumerated. Characteristics of the strain and the matrix are shown in table 13.

2.2.1.2. Spiking protocol
Six levels of contamination were tested including the negative control. Six replicates for each level of contamination were inoculated and analyzed by the reference method and the alternative method. As the two methods have a common step, 6 test portions of 25 g were prepared for each level of contamination and individually inoculated with a calibrated bacterial suspension. Bacterial suspensions of about 10 cells per mL was prepared. From this initial suspension, volumes of 0.9 mL, 0.3 mL and 0.1 mL were used to spike 25 g of sample respectively for the 3 first levels. In parallel, the initial suspension was diluted ratio ½ and ¼ in order to inoculate the lower levels of contamination with 0.1 mL. For all the levels of contamination, homogeneity of the inoculums was checked by enumeration on 30 TSA Petri dishes. Then, the confidence interval was determined according to Poisson law.

2.2.2. Third renewal In order to fulfil the specific requirements of the Technical Board linked to the standard ISO 16140:2003 for the validation of alternative methods, the RLoD was determined for the category “Feedstuff” using the protocols described in the standard ISO/FDIS 16140‐2:2015. Statistical tests and interpretation of the results were still performed according to the standard ISO 16140:2003.
2.2.2.1. Matrix
A couple “matrix‐strain" was studied in parallel with the reference method and the alternative method for the category “Feedstuff”. The total viable count of the matrix was enumerated. Characteristics of the strain and the matrix are shown in table 16.

2.2.2.2. Spiking protocol
Three levels of contamination were tested including the negative control. The negative control level shall not produce positive results. Five replicates are tested for this level. The low level shall be the theoretical detection level, it has been contaminated at 0.7 ‐ 1 CFU per test portion to obtain fractional recovery results. Twenty replicates are tested for this level. The higher level shall be just above the theoretical detection level, it has been contaminated at 2 ‐ 3 CFU per test portion. Five replicates are tested for this level. The matrix was contaminated using the seeding protocol. Bulk contaminations were performed on the matrix for the different levels of contamination, then the matrix was stored at 5±3°C for two days before analysis.

2.3. Inclusivity / exclusivity (selectivity) The objective of this study is to test:
‐the inclusivity: the detection of the target microorganism from a wide range of strains, ‐the exclusivity: the lack of interference from a relevant range of non‐target microorganisms.
According to the requirements of NF EN ISO 16140, 53 strains of Salmonella spp and 30 non‐target strains were tested. A list of the strains figures in appendix 4.
2.3.1. Test protocols
Inclusivity Each Salmonella strain was cultivated twice before inoculation in buffered peptone water (about 1 to 100 CFU/225 mL). The complete protocol of the alternative method was applied with the minimum time of incubation.
Exclusivity Each non‐target strain was cultivated twice before inoculation in growth medium (Tryticase Soy Broth) with a level of contamination expected to occur in the food matrices (about 105 CFU/mL). After 24 hours of incubation, the SMS® test was performed. In cases where the target strains or non‐target strains results were unexpected to interpret by the alternative method, the analysis was conducted again in parallel with the alternative method and the reference method (complete protocol).
2.3.2. Results
Inclusivity 53 Salmonella strains were tested. All strains gave an arc of migration and a red colouration of the medium except for the serovar Paratyphi A, whose strains only gave the arc of migration associated with a low red colouration or an absence of colouration. This is due to this specific serovar of Salmonella whose lysine decarboxylation activity is low or zero. The serovar Gallinarum gave negative results. This serovar is not detected by SMS® test because it is non‐motile. One S. Infantis strain and one S. paratyphi C strain gave negative results. However 3 other strains of Salmonella. Infantis and another strain of Salmonella Paratyphi C gave positive results.
Exclusivity 30 non‐target strains were tested. No cross reaction was observed.

Two strains of Paratyphi A and one strain of Paratyphi C were tested upon request of the Technical Committee. Results were positive for the three strains, despite the absence of coloration observed for the strains of Paratyphi A. Results are shown in appendix 4 bis.
2.3.4. Conclusion The selectivity of the method is satisfactory. No cross reactions was observed with non‐target strains. However non‐motile serovars are not detected.
2.3.5. Cold storage of the SMS Petri dishes A study for the conservation of the SMS Petri dishes was realized during the extension study in 2007 on 30 target strains (see § 5). After storage, the SMS Petri dishes were read and confirmed. The results were consistent with those expected for the 30 strains.
2.4. Conclusion The performances of the SMS test for the detection of Salmonella spp are comparable to those of the method EN ISO 6579. This study concerned 432 samples of six categories of products (meat, dairy, egg, seafood and vegetable, feedstuffs, environmental samples). Values obtained for the 3 criteria are the following:
‐relative accuracy: 98.6% ‐relative sensitivity: 98.6% ‐relative specificity: 98.6%
Six discordant results were observed (3 ND and 3 PD). The relative level of detection of the alternative method and the reference method was evaluated for all categories. The results are comparable because the detection limit of the both methods varies from 0.2 to 0.9 CFU /25 for all categories. The specificity of the method is satisfactory. No cross reactions were observed with non‐target strains. All the tested serovars of Salmonella were detected by the alternative method at the incubation times specified in the protocol, except non motile strains of Salmonella.

3. Interlaboratory study The main object of the interlaboratory study is to determine the variability of the results obtained by different laboratories analysing identical samples and to compare these results within the framework of the comparative study of the methods.
3.1. Interlaboratory study implementation
3.1.1. Participating laboratories The interlaboratory study was realized by the expert laboratory and fourteen participating laboratories.
3.1.2. Salmonella spp absence in the matrix Before spiking, the absence of Salmonella spp was verified in the batch of pasteurized milk used according to the reference method.
3.1.3. Strain stability in the matrix The total viable count (TVC) of several pasteurized milks was enumerated to choose a matrix which contains an annex microflora. The results showed a TVC inferior to 1 CFU/mL for all the matrices analysed. The strain stability in the pasteurized milk matrix was evaluated for 4 days at (4±2)°C. The strain used was Salmonella Enteritidis, a wild strain isolated from an egg product. The detection of Salmonella spp was realized after inoculation of about 10 cells in 25 mL of pasteurized milk. The samples were analyzed at D0, D+1, D+2 and D+3 by reference and alternative methods. The results are summarized in table 18.

The results show that the Salmonella strain used is stable for 3 days at (4±2)°C in the pasteurized milk matrix.
3.1.4. Samples preparation and spiking The matrix was inoculated with the target strain suspension to obtain 3 contamination levels: ‐L0: 0 cell in 25 mL ‐L1: 3 cells in 25 mL ‐L2: 30 cells in 25 mL The matrix was distributed at 25 mL in sterile vials. Every vial was individually spiked and homogenized. Eight samples per level, per laboratory and per method were prepared. Each laboratory received 24 samples to analyse, 1 sample to quantify the endogenous microflora and 1 water sample containing a temperature probe. The results of the enumerations of the TVC, the target levels and the real levels of contamination are presented in table 19.

3.1.5. Samples labeling The labelling of the vials was realized as follows: a code to identify the laboratory: from A to N (cf. table 20) and a code to identify each sample, only known by the expert laboratory. The samples and the temperature control vials (water sample with a temperature probe) were stored at 4°C before shipping.

3.1.6. Samples shipping, reception and analysis The samples were shipped in a coolbox on the 30th of March 2004. The coolboxes were received the 31st of March by all laboratories. The control temperature was recorded upon receipt of the package and the temperature probe sent to the expert laboratory. The samples were analyzed on the 31st of March 2004 by twelve participating laboratories. The expert laboratory concurrently analyzed a set of samples under the same conditions with both methods.

Results analysis Results are consistent with those expected for all laboratories, except for the laboratory D which found only 6 positive on 8 at level L1 and L2 with the alternative method. This laboratory made mistakes in labelling and switched the results. According to this finding, the expert laboratory proposed to exclude the results of laboratory D of the statistical analysis of the results. This proposition was accepted by the Technical Committee. Final analysis was consequently conducted using data supplied by thirteen laboratories.

3.2.7. Discordant results analysis Discordant results are analyzed according to the annex F of ISO 16140 standard. The total number of discordant results is given by the following formula: Y = PD + ND. In the present case, Y = 0, so Y<6, no tests are available. The methods are considered as equivalent.
3.3. Interpretation
3.3.1. Accordance The accordance is the percentage chance of finding the same result (i.e. both negative or both positive) from two identical test portions analysed in the same laboratory, under repeatability conditions (i.e. one operator using the same apparatus and same reagents within the shortest feasible time interval). To derive the accordance from the results of an interlaboratory study, the probability that two samples give the same result is calculated for each participating laboratory in turn, and this probability is then averaged over all laboratories. Values of accordance are shown in table 28. Calculations of accordance by level and method are presented in appendix 6.

3.3.2. Concordance The concordance is the percentage chance of finding the same result for two identical samples analysed in two different laboratories. To calculate the concordance from the results of an interlaboratory study, take in turn each replicate in each participating laboratory, pair it with identical results of all the other laboratories. The concordance is the percentage of all pairings giving the same results on all the possible pairings of data. Values of concordance are shown in table 29. Calculations of concordance by level and method are presented in appendix 7.

3.3.3. Concordance odds ratio If the concordance is smaller than the accordance, it indicates that two identical samples are more likely to give the same result if they are analysed by the same laboratory than if they are analysed by different ones, suggesting that there can be variability in performance between laboratories. Unfortunately, the magnitude of the concordance and accordance is strongly dependent on the level of accuracy, making it difficult to assess easily the degree of between‐laboratory variation.

It is therefore helpful to calculate the concordance odds ratio (COR) defined as follows: COR = accordance x (100‐concordance)/concordance x (100 – accordance). Values of COR for both methods are shown in table 30. A value for the odds ratio of 1.00 would be expected if accordance and concordance were equal, and the larger the odds ratio is, the more inter‐laboratory variation is predominant. Nevertheless, values above 1.00 can occur by chance variation, and so a statistical significance test should be used to confirm whether the evidence for extra variation between laboratories is convincing. The “exact test” is the best recommended test for this). The philosophy behind such tests is that the probabilities of occurrence are calculated for all sets of replicate results that could have produced the overall numbers of positives and negatives.

The values obtained during the interlaboratory study are better than those obtained during the comparative study, probably because of the greater variety of samples and strains tested during the preliminary study. The sensitivity of both methods is recalculated in table 32 by including all confirmed positive results.

3.4. Conclusion The results obtained for the 13 selected laboratories showed that the values of relative accuracy, relative sensitivity and relative specificity are comparable to those obtained during the comparative study. The variability of the alternative method, demonstrated by the calculations of accordance, concordance and concordance odds ratio, is similar to that of the reference method.

5. Extension study An extension study was done in 2007 (documents in appendix 8). This study aimed to add an option for additional confirmation. Tests were made in pure cultures growing on the SMS media and may represent a characteristic profile: 150 strains of Salmonella serotypes from various origins were tested and 105 non‐target strains (the choice being guided by the genetic similarity with Salmonella spp.) Two protocols were tested:
Protocol 5 hours By streaking from a presumptively positive SMS Petri dish in brain heart infusion broth (BHI) and incubated for (5 ± 1)h at (37 ± 1)°C followed by an agglutination test (antigen‐antibody) latex.
Protocol 24 hours By streaking from a presumptively positive SMS Petri dish on SALSA medium and incubated for (21 ± 3)h at (37 ± 1)°C followed by an agglutination test (antigen‐antibody) latex. SALSA medium: This is an agar medium composed of 2 specific and selective media (XLD and ASAP) for Salmonella spp. The medium SALSA is arranged in bi‐Petri dish:
ASAP (white): its mode of action is based on the detection of enzyme activity (C8‐esterase) which specifically cleaves a chromogenic substrate and colours the colonies of Salmonella in pink.
XLD (red): its mode of action is based on the decarboxylation of L‐lysine and / or production of hydrogen sulfide (H2S) giving red colonies with or without black centers. Agglutination test uses latex particles sensitized with rabbit antibodies which agglutinates to Salmonella spp antigens forming aggregates clearly visible.
Results for target strains The results obtained for 144 strains are consistent with those expected. Strains with a positive profile on SMS agar form characteristics colonies on ASAP medium and / or XLD medium and show a positive reaction in the agglutination assay. Seven of 150 strains give a result different than expected: ‐Three strains are self‐agglutinating [S. Cerro (P24), S. salamae (P62) and S. Urbana (P54)]. The latex test, made from colonies obtained on different agar media (ASAP, XLD, TSA), is unusable. The agglutination reaction is positive from the BHI broth (at 5 and 24 h of incubation) for P62 and P54 strains, while the result is negative for the strain P24. Additional testing (confirmatory tests according to standard NF EN ISO 6579) show that it is a strain of Salmonella spp. ‐Three strains [S. Arizonae (P64), S. Braenderup (P58) and S. Diarizonae (P65)] have a negative profile on the SMS medium. The results obtained from the TSA are consistent with those expected (typical colonies on XLD and ASAP and positive reaction in the agglutination assay). ‐The profile of one strain of Salmonella Paratyphi C (R106) is negative on SMS medium. From the colonies obtained on the TSA, R106 does not form characteristic colonies on XLD and the agglutination reaction is positive for the strain regardless of the modality tested. Remarks: ‐Five strains form typical colonies on the ASAP medium and atypical colonies on XLD medium [S. London (P17), S. Montevideo (P25), S. Regent (P53), S. Tennessee (P21) and S. Worthington (P18)]. ‐A strain gives characteristic colonies on XLD medium and atypical colonies on the ASAP medium [S. Dublin (S59)].

‐The profiles of two strains of Salmonella Paratyphi A (R105 and R107) are colorless on SMS medium, due to the absence of lysine decarboxylase activity in these strains. They were indicated negative in the first study because of the absence of coloration which was only considered at this time of the validation. At the request of the Technical Committee, additional tests were performed. For 30 target strains (30 different serotypes), the SMS medium showing a positive profile were stored at (4 ± 2)°C for 48 hours. The analytical protocol (BHI, SALSA and agglutination test) was then applied. The results obtained from an SMS agar positive after storage for 48 hours at (4 ± 2) ° C are identical to results obtained with the general protocol.
Results for non‐target strains The results are consistent with those expected. All strains tested gave a negative profile on SMS agar. No strains formed latex agglutination test, with the exception of a strain of Serratia marcescens (W34). For Serratia marcescens (W34), the reaction is obtained in the form of filamentous aggregates which can be confused with a normal agglutination reaction. Remarks: Some non‐target strains formed characteristic colonies on XLD and ASAP media. ‐Four strains form characteristic colonies on XLD medium [Citrobacter freundii (R35), Citrobacter freundii (W3), Proteus mirabilis (W29) and Proteus mirabilis (W30)]. ‐Two strains form characteristic colonies on the ASAP medium [Enterobacter sakazakii (I37) and Pseudomonas fluorescens (R4)]. However, the latex agglutination tests performed on typical colonies formed by these strains are negative.
Conclusion Case No. 2 proposed for confirmation of presumptive positive test results by SMS confirmation method is specific for strains of Salmonella spp. The result of the strain S. Cerro (P24) is unusable for the agglutination test as it is a self‐agglutinating strain regardless of the modality tested. The results obtained with three target strains showed a mismatch between the test SMS and the confirmatory test. Indeed, the three strains [S. Arizonae (P64), S. Braenderup (P58) and S. Diarizonae (P65)] give a negative profile of the SMS agar while testing (SALSA and agglutination test) made from the TSA agar give positive results. However, other strains of the same serotypes (Arizonae, Braenderup and Diarizonae) give a positive profile on SMS agar.

6. Conclusion
The performances of the SMS test for the detection of Salmonella spp are comparable to those of the method EN ISO 6579. This study concerned 432 samples of six categories of products (meat, dairy, egg, seafood and vegetable, feedstuffs, environmental samples).
Values obtained for the 3 criteria are the following: ‐relative accuracy: 98.6% ‐relative sensitivity: 98.6% ‐relative specificity: 98.6%
Six discordant results were observed (3 ND and 3 PD).
The relative level of detection of the alternative method and the reference method was evaluated for all categories. The results are comparable because the detection limit of the both methods varies from 0.2 to 0.9 CFU /25 for all categories.
The specificity of the method is satisfactory. No cross reactions were observed with non‐target strains. All the tested serovars of Salmonella were detected by the alternative method at the incubation times specified in the protocol, except non‐motile strains of Salmonella.
The test proposed for confirmation of presumptive positive test results by SMS confirmation method is specific for strains of Salmonella spp.
The results obtained in interlaboratory study for the 13 selected laboratories showed that the values of relative accuracy, relative sensitivity and relative specificity are comparable to those obtained during the comparative study. The variability of the alternative method, demonstrated by the calculations of accordance, concordance and concordance odds ratio, is similar to that of the reference method.
