BIOCATALYST In addition to structural proteins, cells also synthesize a group of proteins that have a
specific function enzymes as biocatalyst. By enzyme biocatalyst is a very important aspect in
biochemical reactions in the cell bodies of living. Almost all biochemical reactions in the cell are
catalyzed by enzymes. The enzyme has the ability to accelerate the transformation of the chemical
reaction at least to a million-fold when compared with the chemical transformation without a
catalyst. However, the enzyme did not change the balance of the reaction, but rather lowers the
activation energy of a reaction. The enzyme has the main characteristics of the catalytic ability and
specificity. Almost all enzymes known form of the protein, although there are RNA molecules that
also have catalytic properties (ribozymes), for example RNAseP a role in tRNA processing in
Escherichia coli, and the RNA that play a role in the processing of rRNA in Tetrahymena.
BIOKATALIS
Selain protein struktural, sel juga menyintesis sekelompok protein yang mempunyai fungsi
khusus yaitu enzim sebagai biokatalis. Biokatalis oleh enzim merupakan aspek yang sangat penting
dalam reaksi biokimia di dalam sel jasad hidup. Hampir semua reaksi biokimia di dalam sel dikatalisis
oleh enzim. Enzim mempunyai kemampuan mempercepat reaksi transformasi kimiawi paling tidak
sampai sejuta kali lipat jika dibandingkan dengan transformasi kimia tanpa katalis. Meskipun
demikian, enzim tidak mengubah keseimbangan reaksi , melainkan menurunkan energi aktifasi suatu
reaksi. Enzim mempunyai karakteristik utama yaitu kemampuan katalitik dan spesifitas. Hampir
semua enzim yang telah diketahui berupa protein, meskipun ada molekul RNA yang juga
mempunyai sifat katalitik (ribozim), misalnya RNAseP yang berperan dalam pemrosesan tRNA pada
Escherichia coli , dan RNA yang berperan dalam pemrosesan rRNA pada Tetrahymena.

mempunyai keterkaitan yang sangat dekat. Reaksi samping yang mengarah ke pembentukan produk
sampingan biasanya tidak terjadi pada reaksi yang dikatalisis oleh enzim. Selain itu spesifisitas enzim
terhadap substrat umumnya sangat tinggi bahkan seringkali mutlak, artinya satu macam enzim
hanya akan bereaksi terhadap satu macam substrat. Sebagai contoh, enzim proteolitik hanya akan
menghidrolisis suatu ikatan peptida. Tripsin adalah salah satu contoh enzim proteolitik yang
mengkatalisis pemutusan ikatan peptida hanya pada sisi karboksil residu lisin dan arginin. Thrombin
adalah enzim yang bahkan lebih spesifik daripada tripsin karena menghidrolisis ikatan arginin – lisin
hanya pada urutan peptida tertentu.

Meskipun enzim mempunyai kemampuan mempercepat reaksi, namun kemampuan katalitik
tersebut tidak secara terus menerus diaktifkan. Banyak cntoh enzim yang aktivitasnya dikendalikan
dengan mekanisme tertentu. Enzim yang mengkatalisis reaksi pertama dalam suatu jalur biosintetik
biasanya akan dihambat aktifitasnya oleh hasil akhir jalur reaksi tersebut (penghambatan umpan
balik, feed- back inhibition). Sebagai contoh, biosintesis isoleusin pada bakteri mempunyai sistem
pengendalian aktivitas enzim semacam ini. Enzim pertama yang berperan didalam biosintesis
isoleusin, yaitu treonin deaminase, akan dihambat aktivitasnya jika konsentrasi isoleusin di dalam sel
sudah mencapai batas tertentu. Skema penghambatan umpan balik suatu enzim digambarkan pada

Growth of microorganisms
Growth can be defined as all components are added on a regular basis within the living cell.
1. In multicellular organisms

Pertumbuhan jasad renik
Pertumbuhan dapat didefinisikan sebagai pertambahan secara teratur semua komponen di dalam
sel hidup.
1. Pada Organisme Multiseluler
Adalah peningkatan jumlah sel per organisme, dimana ukuran sel menjadi besar.
2. Pada Organisme Uniseluler
Adalah pertambahan jumlah sel, yang berarti juga pertambahan jumlah organisme, misalnya
pertumbuhan yang terjadi pada suatu kultur mikroorganisme.
3. Pada Organisme Soenositik/Aseluler
Selama pertumbuhan, ukuran sel menjadi bertambah besar tetapi tidak terjadi pembelahan
sel.
Pertumbuhan disebut dalam keadaan keseimbangan jika terjadi secara teratur dalam
keadaan konstan, sehingga jumlah pertambahan komponen kimia juga konstan. Sebagai contoh
pertambahan jumlah masa sel sebanyak dua kali dalam keadaan keseimbangan akan mengakibatkan
pertambahan jumlah komponen sel seperti air, protein, RNA, DNA dan sebagainya sebanyak dua kali
pula.
Umur sel ditentukan segera setelah proses pembelahan sel selesai, sedangkan umur kultur
ditentukan dari waktu atau lamanya inkubasi. Ukuran sel tergantung dari kecepatan
pertumbuhannya. Semakin baik zat nutrisi di dalam substrat tempat tumbuhnya, mengakibatkan
pertumbuhan sel semakin cepat dan ukuran sel semakin besar. Pertumbuhan sel bakteri, kapang dan
khamir akan dibahas pada masing-masing bab mengenai bakteri, kapang dan khamir.

1. Adaptation phase

medium tempat tumbuhnya., seperti : pH, kandungan nutrien, kondisi lingkungan (suhu dan
kelembaban udara). Pada fase ini, sel mikroorganisme membutuhkan energi lebih banyak dari pada
fase lain. Selain itu sel paling sensitif terhadap keadaan lingkungan.
Phase Slow Growth / Reduction Growth phase (Phase of Negative Growth Acceleration)

Pada fase ini pertumbuhan jasad renik diperlambat karena beberapa sebab :
a. Zat nutrisi di dalam medium sudah sangat berkurang
b. Adanya hasil-hasil metabolisme yang mungkin beracun dapat menghambat pertumbuhan
jasad renik.
Pada fase ini, pertumbuhan sel tidak stabil tetapi jumlah populasi masih naik karena jumlah
sel yang tumbuh lebih besar dari jumlah sel yang mati.
2. Stationary Phase / Phase constant (Maximum Stationary Phase)

sama/berbeda dengan sel pada fase logaritmik. Pada fase ini, sel-sel menjadi lebih tahan terhadap
keadaan ekstrem, seperti : panas, dingin, radiasi, bahan kimia.

FACTORS - FACTORS AFFECTING GROWTH MICROORGANISMS

FAKTOR - FAKTOR YANG MEMPENGARUHI PERTUMBUHAN JASAD RENIK
Faktor-faktor yang mempengaruhi pertumbuhan jasad renik yang bersifat heterotrof adalah
tersedianya nutrien, air, suhu, pH, oksigen dan potensial oksidasi-reduksi, adanya zat penghambat
dan adanya jasad renik lain.
1. NUTRIENT
Microorganisms need nutrients for life and growth, namely as:

c. energy sources
d. Growth factors are minerals and vitamins

Jasad renik membutuhkan nutrien untuk kehidupan dan pertumbuhannya, yaitu sebagai :

berbeda sehingga solut (nutrien) bisa masuk dan terjadi keseimbangan konsentrasi terutama
pada nutrien dengan berat molekul kecil. Sebagai contoh, misalnya air yang dapat keluar
masuk sel secara bebas.

pertumbuhan sel jasad renik di dalam suatu makanan sangat dipengaruhi oleh jumlah air yang
tersedia. Selain merupakan bagian terbesar dari komponen sel (70 – 80 %), air juga dibutuhkan
sebagai reaktan berbagai reaksi biokimia. Tidak semua air yang terdapat dalam bahan pangan dapat
digunakan jasad renik.
Beberapa kondisi atau keadaan dimana air tidak bisa digunakan oleh jasad renik:
Adanya solut dan ion yang dapat mengikat air didalam larutan
Misalnya : gula dan garam konsentrasi tinggi akan mengikat air dari bahan pangan, bahkan dapat
mengikat air dari dalam sel jasad renik jika konsentrasi solut diluar sel lebih besar daripada di dlm
sel.
Koloid hidrofilik (gel) dapat mengikat air sebanyak 3-4 % agar dapat menghambat
pertumbuhan bakteri didalam medium
Air dalam bentuk kristal es atau hidrasi tidak dapat digunakan oleh jasad renik
PH VALUE
Medium pH value greatly affects the types of microorganisms that can grow.

from 6.5 to 7.5 and can not grow well at a pH below 5.0 and a pH above 8.5 except for acetic acid

Yeasts can grow at a pH of 2.5 to 8.5 and pH optimum growth was at pH 4-5. Therefore, the
yeast growth at low pH where the bacterial growth is inhibited. Mold can grow at pH 3 - 8.5 and pH

NILAI PH
Nilai pH medium sangat mempengaruhi jenis jasad renik yang dapat tumbuh. Jasad renik
pada umumnya dapat tumbuh pada kisaran pH 3 – 6. Kebanyakan bakteri mempunyai pH optimum
pertumbuhan 6,5 – 7,5 dan tidak dapat tumbuh baik pada pH dibawah 5,0 dan pH diatas 8,5 kecuali
bakteri asam asetat (Acetobacter suboxydans), serta bakteri oksidasi sulfur.
Khamir dapat tumbuh pada pH 2,5 – 8,5 dan pH optimum pertumbuhan adalah pada pH 4 –
5. Oleh karena itu, khamir tumbuh pada pH rendah dimana pertumbuhan bakteri terhambat. Kapang
dapat tumbuh pada pH 3 – 8,5 dan pH optimum pertumbuhan 5 – 7 .

Makanan dengan pH rendah (dibawah 4,5), biasanya tidak ditumbuhi oleh bakteri, tetapi
dapat menjadi rusak karena pertumbuhan khamir dan kapang. Oleh karena itu, makanan yang
mempunyai pH rendah relatif lebih tahan selama penyimpanan dibandingkan dengan makanan yang
mempunyai pH netral atau mendekati netral.
Dalam pengolahan pangan, makanan dapat dibedakan atas beberapa gruo berdasarkan pH
nya. Pembagian makanan atas beberapa grup ini bertujuan untuk mengetahui daya awet suatu
makanan. Dengan demikian, memudahkan mencari perlakuan yang harus diberikan untuk
mengawetkan makanan tersebut. Semakin rendah pH makanan, semakin berkurang perlakuan
pengawetan makanan yang harus diberikan pada makanan tersebut.

maksimum pertumbuhannya maka aktivitas enzim akan berhenti, bahkan pada suhu yang terlalu
tinggi akan terjadi denaturasi enzim. Jasad renik dapat dibedakan atas beberapa grup berdasarkan
atas kemampuannya untuk dapat memulai pertumbuhan pada kisaran suhu tertentu.
Suhu penyimpanan bahan makanan sangat berpengaruh besar terhadap jenis dan kecepatan
pertumbuhan jasad renik. Kapang dan khamir pada umumnya merupakan golongan mesofil, dapat
tumbuh baik pada suhu 25 – 30 oC. Oleh karena itu, tumbuh dengan baik pada makanan yang
disimpan pada suhu kamar, bahkan beberapa masih dapat tumbuh pada suhu pendinginan.

Pertumbuhan jasad renik terjadi pada suhu dengan kisaran (antara suhu minimal dan
maksimal) kira-kira 30 0C.
Kecepatan pertumbuhan jasad renik meningkat lambat dengan naiknya suhu sampai
mencapai kecepatan pertumbuhan maksimum.
Diatas suhu maksimum, kecepatan pertumbuhan menurun dengan cepat dengan naiknya
suhu.

Konsentrasi oksigendi dalam bahan makanan dan lingkungannya berpengaruh terhadap jenis
jasad renik yang tumbuh pada makanan tersebut.

Anaerobik (tidak membutuhkan oksigen)
