• 8 PCR reactions for the detection of HLA-B*51 (associated with Morbus Behçet), HLA-B*57 (associated with HSR to Abacavir).

• Behçet disease (Behçet‘s syndrome, Morbus Behçet, silk road disease) is associated among other things with HLA-B*51 and a form of vasculitis that can lead to ulceration and other lesions. The resulting infl ammation affects blood vessels, usually the small ones. Therefore symptoms occur wherever there is a patch of infl ammation, and can be anywhere where there is a blood supply.

• Only 1 PCR reaction for DQB1*06:02, ideal for screening patients for Narcolepsy.

• Narcolepsy („sleeping sickness“, „slumber addiction“) belongs to the group of sleep disorders (dyssomnia), which are based on a neurological disorder of the sleep- wake rhythm. The cause of the disorder is a selective loss of neurons of the perifornical hypothalamus which produce hypocretins (orexins).

• Only 2 PCR reactions for screening DQB1*02, *03:02 and DQA1*05:01, *05:05.

• Coeliac Disease is one of the most frequent chronic diseases of the small intestine caused by an intolerance against gluten (protein in diverse types of grains like wheat, rye, barley,... ).

• Only one PCR reaction, ideal for screening patients for B*57. • Many HIV infected patients showing a hypersensitivity reaction (HSR) to Abacavir are carrier of the HLA-B*57:01 allele.

• Killer-cell immunoglobulin-like receptors (KIRs) are a family of cell surface proteins found mainly on natural killer (NK) cells. They regulate the killing function of these cells by interacting with HLA class I molecules, which are expressed on all cell types.

• Most KIRs are inhibitory, meaning that their recognition of HLA suppresses the cytotoxic activity of their NK cell.

• The Ready Gene Wipe Test enables the user to monitor for potential contaminations with amplicons created by the Ready Gene product line on working areas as required by International Standards for Histocompatibility Testing (e.g. ASHI/EFI).

HLA-Ready Gene B*15:02 - HYPERSENSITIVITY REACTION (HSR) TO CARBAMAZEPINE

• Only one PCR reaction, ideal for screening patients for A*31. • Carbamazepine is commonly prescribed for the treatment of epilepsy, trigeminal neuralgia and bipolar disorders.• The HLA-A*31:01 allele is strongly associated with Carbamazepine-induced hypersensitivity syndrome in Northern European patients. Furthermore it is a risk factor for Carbamazepine-induced maculopapular exanthema in Han Chinese populations and Stevens- Johnson Syndrome and toxic epidermal necrolysis (SJS-TEN) in patients from Japan and from Northern Europe.

• Even though the prescence of the HLA-A*31:01 allele is neither necessary nor suffi cient for the development of a HSR to Carbamazepine, it is correlated with a signifi cantly increased risk.• With a negative A*31 reaction the risk of a HSR is decreased.• McCormack et al.: HLA-A*3101 and Carbamazepine- induced hypersensitivity reactions in Europeans, N Engl J Med 2011; 364: 1134-43.

• Only 2 PCR reactions for screening patients for B*15:02. • The HLA-B*15:02 allele has been shown to be strongly correlated with Carbamazepine-induced Stevens-Johnson Syndrome and toxic epidermal necrolysis (SJS-TEN) in Asian populations, i.e. Han Chinese, Malay, Thai and Indians.•Carbamazepine is the most common cause of SJS-TEN in South-east Asian countries.• Among Han Chinese individuals, Carbamazepine-induced SJS-TEN almost never occurs in noncarriers of the HLA-B*15:02 allele.

• With negative HLA-B*15:02 reactions the risk of Carbamazepine-induced SJS-TEN can be excluded.• The FDA recommends the HLA-B*15:02 genotyping of patients of Asian ancestry before initiating Carbamazepine therapy.• Chen et al.: Carbamazepine-induced toxic effects and HLA-B*1502 screening in Taiwan, N Engl J Med 2011; 364: 1126-33.
