Theory and Function
Photon Correlation Spectroscopy
Electrophoretic light scattering is the method mostpopularly used to determine the velocity of theparticles suspended in a fluid medium underan applied electric field. In order to determine thespeed of the particles movement, the particles areirradiated with a laser light and the scattered lightemitted from the particles is detected. Since thefrequency of the scattered light is shifted from theincident light in proportion to the speed of theparticles movement, the electrophoretic mobility
of the particles can be measured from the frequencyshift (Doppler shift) of the scattered light.
Measurement Process
When an electric field is applied to charged particlesin the suspension, particles move toward an electrodeopposite to their surface charge. Since the velocity isproportional to the amount of charge of the particles,zeta potential can be estimated by measuring thevelocity of the particles.
Photon correlation spectroscopy is a techniqueused to determine the diffusion coefficient of smallparticles in fluid. The coefficient is determinedby accurately measuring the light scattering intensityof the particles as a function of time.
Measurement Process
As the particles of interest diffuse within the samplecell due to Brownian motion, an incident beam oflaser light illuminates the particles. The particles scatterthe light, producing fluctuations in the scatteringintensity as a function of time. The scattered lightis collected at a chosen angle, and is measuredby a highly sensitive detector. Since the diffusion rate
of particles is determined by their size, informationabout their size is contained in the rate of fluctuationof the scattered light. So, by correlating the fluctuation,we can determine the particle size distribution of thepopulation present.
One of the Widest Concentration Ranges
The DelsaNano is capable of analyzing samples withconcentrations ranging from 0.001% to 40% (sampledependent), for both particle size and zeta potential.
Zeta Potential of Concentrated Samples
Electrophoretic light scattering measurementson concentrated samples are now available, thanksto the DelsaNano patented Forward Scatteringthrough Transparent Electrode Technology (FST).This method detects the scattered light from particlesthrough a transparent electrode at a 30° scatteringangle by using a short optical path that reduces effectsdue to multiple scattering with very high accuracy.
